ABSTRACT

Objective To prepare 4-sulfonylcalix[6]arene-modified cotton fibers for adsorption and removal of uranium based on the specific complexation of calix[6]arene with uranium (VI). Methods Chemical grafting was used for the modification of cotton, which reacted with α-bromoisobutyryl bromide, glycidyl methacrylate, and 4-sulfonylcalix[6]arene. Scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and infrared spectroscopy (FTIR) were used to characterize the structure of 4-sulfonylcalix[6]arene-modified cotton (Cotton S-C[6]a). A Franz diffusion cell was used to simulate uranium-contaminated skin. Laser fluorimetry was used to determine the uranium content. Results SEM, XPS, and FTIR showed that cotton fibers were successfully grafted with 4-sulfonylcalix[6]arene. The optimal conditions of Cotton S-C[6]a for the adsorption of uranium (VI) was pH 4.0, duration of 20 min, and 20 mg of adsorbent. The adsorption process fitted well with pseudo-secondary-order kinetics. The uranium removal efficiency of Cotton S-C[6]a was up to 78.46% in aqueous solution and 81.72% on skin. Conclusion The synthesized Cotton S-C[6]a is highly efficient in the removal of uranium (VI) in solution and on contaminated skin.

ABSTRACT

Objective:To explore the disturbance of metal element balance in mice after exposure to radon.Methods:Mice were randomly divided into control group, radon exposure of 30 WLM group, 60 WLM group and 120 WLM groups, with 10 mice in each group. After radon exposure with the cumulative dose, the lung function of mice was detected by a non-invasive pulmonary function testing instrument. Mice blood was taken from eyeballs. The lungs, heart, liver, kidney and spleen were also collected. HE staining was used to observe the pathological changes of lung tissue. Inductively coupled plasma mass spectrometry (ICP-MS) was used to detect the content of metal elements, including essential trace elements in the body: chromium (Cr), molybdenum (Mo), cobalt (Co), selenium (Se), copper (Cu), zinc (Zn), manganese (Mn), nickel (Ni), and potentially toxic elements: arsenic (As), tin (Sn), lead (Pb), aluminum (Al), mercury (Hg), cadmium (Cd), and silver (Ag).Results:Compared with the control group, lung ventilation function of the radon-exposed mice was decreased, alveolar structure was destroyed, and the contents of pulmonary metal elements Cr, Al, Pb, Sn( F=0.34, 0.66, 3.14, 1.16, P<0.05) and essential trace elements Mn, Cr, Zn, and Mo in the blood were decreased( F=0.65, 1.44, 0.97, 2.08, P<0.05), while the elements of Cu, Mo, Se and As in the lungs were increased( F=1.31, 1.26, 0.81, 2.04, P<0.05), and the element contents in other tissues also fluctuated. Conclusions:Inhalation of a certain cumulative dose of radon can reduce the lung ventilation function of mice and induce lung inflammation, as well reduce the content of essential trace elements in the lung and blood so that the content of metal elements in the body fluctuates.

@#<b>Objective</b> To investigate the role of complement in radiation-induced lung injury in mice after chest irradiation with <sup>60</sup>Co γ-rays at a single dose of 20 Gy. <b>Methods</b> C57BL/6 mice underwent chest irradiation with <sup>60</sup>Co γ-rays at a single dose of 20 Gy, followed by observation for the inflammatory reaction of the lung tissue in the early stage (within 15 d) and pulmonary fibrosis in the later stage (30 and 180 d). Enzyme-linked immunosorbent assay was used to measure the levels of C2, C3a, C4, and C5b-9 in the lung tissues at 1, 3, 7, 15, 30, and 180 d after irradiation. The expression of complement mRNA in BEAS-2B cells after irradiation was determined using RT-PCR. <b>Results</b> Radiation-induced lung injury in micepresented as inflammatory response in the early stage and fibrosis in the late stage. Complement C2, C4, and C5b-9 complexes were increased in the early period (3 or 7 d) after irradiation (<i>P</i> < 0.05), which might be associated with the inflammatory response induced by irradiation. During 3 to 180 d, complement C3a was significantly higher in the irradiated mice than in the control mice, suggesting a close relationship between C3a and radiation-induced lung injury. The irradiated cells showed increased mRNA expression of C2 and C3, with no changes in the mRNA levels of C4 and C5. <b>Conclusion</b> Different complement proteins have varying responses to radiation-induced lung injury, among which C3a is closely related to radiation-induced lung injury, suggesting that regulating C3a and its receptors may be a new way to prevent and treat radiation-induced lung injury.

ABSTRACT

@#<b>Objective</b> To explore dendritic cells (DCs)-mediated antigen presentation for radiation-injured cells by using the <i>in vitro</i> cell co-culture technology to simulate the <i>in vivo </i>microenvironment of the lung tissue. <b>Methods</b> <sup>60</sup>Co γ-irradiated mouse lung epithelial cells (MLE-12) were cultured with bone marrow-derived DCs and/or splenic T lymphocytes for 48 hours. Flow cytometry was used to measure the expression levels of costimulatory molecules (CD80/86) and antigenic peptide recognition complexes (the major histocompatibility complex [MHC] class Ⅰ/Ⅱ) on DCs and T cell activation markers (CD69/28/152) as well as the numbers of CD4<sup>+</sup> and CD8<sup>+</sup> T cells. <b>Results</b> <sup>60</sup>Co γ irradiation significantly increased the apoptosis rate of MLE-12 cells in a dose-dependent manner, and significantly stimulated the expression of CD80/86 and MHC Ⅱ on DCs, without direct activation of T cells. After γ (6 Gy)-irradiated MLE-12 cells were co-cultured with DCs and T lymphocytes for 48 h, there were significant increases in the expression of CD69 and CD28 on T cells, the numbers of CD4<sup>+</sup> and CD8<sup>+</sup> T cells, and the expression of CD86 and MHC I on DCs, as compared with the control groups. <b>Conclusion</b> Radiation-injured cells can stimulate antigen presentation by DCs and activate T cells.

ABSTRACT

Objective:To investigate the protective effects of bovine lactoferrin (bLF) supplementation on intestinal mucosal tissue and its influence on of inflammatory factors in the premature rats model of necrotizing enterocolitis(NEC), and to provide the theoretical basis for prevention of NEC by bLF supplementation.Methods:Premature SD rats were randomly divided into 4 groups, 25 cases in each group.Control group: oral feeding; model group : oral feeding with lipopolysaccharides(LPS) gavage + hypoxic stimulation; high dose bLF intervention group: daily bLF (7 g/L) + oral feeding with LPS gavage + hypoxic stimulation; low dose bLF intervention group: daily bLF (2 g/L) + oral feeding with LPS gavage + hypoxic stimulation.Histopathological analysis was performed by HE staining.The expression levels of interleukin-1β(IL-1β)and interleukin-6(IL-6)in intestinal mucosa were detected by enzyme linked immunosorbent assay (ELISA).Results:(1) Morphological observation: the intestinal wall of model group was thin, and there were different degrees of pneumoconiosis and effusion in intestinal cavity.Under the microscopy, it could be observed that the intestinal tissue necrosis was serious, the intestinal villi fell off, glands arranged disorderly, epithelial edema was significant, the lamina propria and submucosa had severely edema and were separated, and there were a large number of inflammatory cells infiltrated.The above-mentioned manifestations were alleviated in the high-dose and low-dose bLF intervention groups, and no significant abnormalities were found in the control group.(2) The expression of IL-1β and IL-6 in intestinal tissue: the tissue concentration of IL-1β and IL-6 in the model group rats [(380.89±20.25) ng/L, (485.12±31.44) ng/L]were significantly higher than those in the control group[(270.69±45.58) ng/L, (212.62±89.46) ng/L]( q =9.785, 14.030, all P<0.01). The expression of IL-1β and IL-6 in mucosal tissue of ileum was significantly inhibited in hypoxic and LPS-stimulated rats fed with bLF(IL-1β: q=9.105, 8.761, all P<0.01; IL-6: q=8.175, 8.996, all P<0.01). There was no significant difference in the expression of IL-1β and IL-6 between high dose bLF(7 g/L) and low dose bLF (2 g/L) inter vention groups (IL-1β: q=-0.084, P>0.05; IL-6: q=-1.140, P>0.05). Conclusion:Enteral bLF supplementation can alleviate the damage of intestinal tissue in NEC model of premature SD rats, inhibit the expression of IL-1β and IL-6 inflammatory factors in intestinal tissue, and have a protective effect on intestinal tissue.

ABSTRACT

Objective@#To investigate the effect of hypertensive disorder complicating pregnancy (HDCP) on the mortality and early complications of premature infants.@*Methods@#The general clinical data of preterm infants with gestational age 24-36+ 6 weeks were collected from the cooperative units in the task group from January 1, 2013 to December 31, 2014.According to the severity of HDCP, the infants were divided into 4 groups: HDCP group, preeclampsia group, eclampsia group and non HDCP group, the mortality and major complications of preterm infants were compared, and the influencing factors were analyzed.@*Results@#The mortality rate of preterm in the HDCP group was significantly higher than that of non HDCP group, and there was statistical significance (χ2=9.970, P=0.019). Eclampsia had a highest fatality rate (4.8%) in the early stage, compared with non HDCP group (2.2%), and the difference was statistically significant.Comparison of HDCP group (1.8%) and eclampsia group (3.2%) suggested that there was no statistically significant difference.The incidence of respiratory distress syndrome (RDS) in preterm in HDCP group was significantly higher than that of non HDCP group, and there was statistical significance (χ2=13.241, P=0.004). Eclampsia group showed the highest incidence (35.4%), compared with non HDCP group (16.2%), the difference was statistically significant, but compared with HDCP group (19.9%), preeclampsia group (17.1%), there was no significant diffe-rence.The incidence of bronchopulmonary dysplasia (BPD) in preterm in HDCP group was significantly higher than that of non HDCP group (χ2=9.592, P=0.022), the highest incidence showed up in eclampsia group (9.7%), compared with non HDCP group (2.0%) and HDCP group (1.7%), the difference was statistically significant.But there was no statistically significant difference, compared with preeclampsia group.As the degree of HDCP aggravated, the incidence of BPD gradually rose.There was no significant impact on necrotizing enterocolitis (NEC), retinopathy of prematurity (ROP), intraventricular hemorrhage (IVH) and sepsis of HDCP (χ2=7.054, 7.214, 0.358, 3.852; P=0.070, 0.065, 0.949, 0.278). Considering the overall outcome of the child, that was, whether the child died or survived, he had at least one complication, and HDCP had an effect on it (χ2=15.697, P=0.001), so the incidence increased while the degree of HDCP rose gradually.After adjusting gestational age, birth weight, sex, way of delivery, placental abruption and front placenta, prenatal hormonal, gestational diabetes, neonatal asphyxia and other factors, the results displayed that HDCP was the factor leading to the death of premature baby (OR=2.159, 95%CI: 1.093-4.266), and comparison between preeclampsia and eclampsia showed no statistical difference (P=0.714, 0.389); HDCP had no significant influence on RDS, BDP, ICH, NEC, ROP and sepsis.@*Conclusions@#HDCP leads to increased risk of premature death, but also leads to the increased incidence of RDS and BPD, but it had no obvious effect on NEC, ROP, IVH, sepsis and other complications.

ABSTRACT

Objective To explore the practice and efficacy of multidisciplinary collaborative management in the prevention and control of multidrug-resistant organism(MDRO)healthcare-associated infection(HAI).Methods MDROs isolated from clinical specimens and MDRO infection in hospitalized patients in April 2012-March 2013(before intervention)and April 2013-March 2014(after intervention)were analyzed retrospectively.Since April 2013,multidisciplinary collaborative management has been implemented,isolation of MDROs and MDRO HAI in hospitalized patients before and after intervention were compared and analyzed.Results 798 and 833 MDRO strains were isolated before and after the intervention respectively,isolation rate of MDROs after intervention was lower than that before intervention(25.71%vs 31.89%,P<0.001).After the implementation of multidisciplinary collaborative management,incidence of MDRO HAI decreased from 0.94%before intervention to 0.63%;hand hygiene compliance rate of health care workers(HCWs)increased from 36.44%before intervention to 53.51%;compliance rate of contact isolation increased from 65.29%before intervention to 90.88%;rational usage rate of antimicrobial agents increased from 64.93%before intervention to 72.53%;specimen detection rate in patients with therapeutic antimicrobial use increased from 41.36%before intervention to 58.72%,differences were all statistically significant(all P<0.001).Conclusion The practice of multidisciplinary collaborative management can effectively implement the prevention and control measures of MDRO HAI,effectively reduce the occurrence of MDRO HAI.

ABSTRACT

Objective To explore the deep venous thrombosis risk assessment table application in the evaluation of patients with respiratory diseases of deep venous thrombosis. Methods Caprini risk score model and Padua score prediction based on the related literature and respiratory diseases in reference to the characteristics of deep venous thrombosis risk factors for deep vein thrombosis, risk assessment table. And in January 2015 October 155 cases of hospitalized patients were evaluated using this table, and all patients underwent lower limb ultrasonography. Results The results showed the risk and the population under ultrasound in 60 cases (score=2 points) thrombosis probability is 0%, 95 cases of high risk population and above (score more than 3 cent) thrombosis probability is 25.26%. There were significant differences between the two groups (χ2=17.94, all P<0.05). Conclusion The application ofnon operative patients with deep venous thrombosis risk assessment table can be a good screening of high-risk patients with thrombosis. For thrombosis in high-risk groups to physical intervention, so as to reduce the incidence of deep venous thrombosis.

ABSTRACT

Objective To analyze the causes of time consumption in CT enhancement scanning (CTES) and CT angiography (CTA) examinations in order to optimize the procedures, and help to save time and medical costs for patients. Methods A total of 2328 outpatients and 1402 inpatients to take CTES and CTA examinations were randomly selected as the normal control group, and another 2085 outpatients and 793 inpatients who underwent the optimized procedures were randomly selected as the experimental group. The problems of time consumption and patients'satisfaction degree were analyzed. Results The major causes for time consumption in CTES and CTA examinations included taking wrong contrast medicine, forgetting to take contrast medicine, having no auxiliary examination results, waiting in the wrong line, and opening the cap of contrast medicine. The time spent for checkup for inpatients and outpatients in the control and experimental groups was (119.8±15.6) minutes and (31.5±8.6 ) minutes vs (55.2 ± 10.6) minutes and (8.4 ±2.1) minutes. The satisfaction degree of inpatients and outpatients in the control and experimental groups were 90.16%(1264/1402) and 88.66%(2064/2328) vs 98.49%(781/793) and 97.94%(2042/2085). The experimental group spent shorter time and had higher satisfaction degree than those in the control group, and the differences were statistical significantly, tinpatient=34.96, P<0.01, toutpatient=12.03, P<0.01;χ2inpatient=55.20, P<0.01,χ2outpatient=146.27, P<0.01. Conclusions After the procedures of CTES and CTA examinations are optimized, the checkup time is significantly shortened, and patients' satisfaction degree is remarkably improved.

ABSTRACT

Necrotizing enterocolitis(NEC)in neonates is the most important neonatal problems in nursery and is associated with high neonatal mortality and morbidity.NEC also leads to various long term complications,including short bowel syndrome,systemic infection,eye problems,nutritional deficiency and neurodevelopmental impairments.Lactoferrin(LF) is a component of breast milk and has multiple actions include antimicrobial,antiviral,anti-fungal and enhance immunity and so on.Studies have been completed and a number of trails are going on for evaluation of efficacy and safety of LF in the prevention of NEC.LF prophylaxis and therapy may have a significant impact on improving clinical outcomes of vulnerable premature infants.

ABSTRACT

Purpose To investigate the diagnostic value of gray-scale ultrasound on ovarian cystic teratoma.Materials and Methods 83 patients with primary diagnosis of ovarian cystic teratoma underwent gray-scale ultrasonography. The ultrasonic images were compared with postoperative pathologic results. Results 91 cases of 83 lesions were detected. The ultrasonic features were categorized into 6 types: cystoid, solid-like, intracystic dough sign, intracystic hair-ball, intracytic fat-fluid level, and intracytic cluttering sign. The diagnose accordance rate for each sign was 81.0%, 77.8%, 96.2%, 100% and 70.0% respectively. The diagnose accordance rate of ultrasonic diagnosis compared with pathologic results was 89.0% (81/91), and misdiagnosed rate and rate of missing diagnosis was 11.0% (10/91). Conclusion The ultrasonic features of ovarian cystic mature terotoma were of importance diagnostic value, whereas differentiation with ovarian chocolate cysts and ovarian mucus-cystic adenoma should be made cautiously.

ABSTRACT

Mastoparan(MAS) and α-latrotoxin(α-LTX) are two kinds of insulinotropic peptides obtained from insect toxins which can interact with islet β-cells and induce insulin secretion. The signal mechanism of these insulinotropic peptides regulating insulin-releasing attracts notable attention and has been elucidated more and more details. MAS mainly acts on the molecular components of exocytosis at a late stage. Insulin secretion induced by MAS is obviously dependent on GTP, which subsequently activates G-protein located on insulin secretion granules(ISG), or activates the Rho subfamily of small G proteins to evoke exocytosis and sensitize fusion machinery. The MAS stimulated insulin-releasing activity can be augmented by nutrients. However, its effect is not Ca~(2+) dependent. There are two regulatory pathway triggered by α-LTX: one way is pore formation caused through plasma membrane, another way is the transmembrane signal transduction evoked by cytosolic second messengers. Tetrameri complexs assembled at high concentration of α-LTX toxin or in the presence of extracellular Ca~(2+), can insert α-LTX into plasma membrane to form Ca~(2+) permeable channels and trigger Ca~(2+)-dependent secretion. By binding to transmembrane receptors and activating phospholipase C, α-LTX induces the generation of second messenger DAG and IP3. IP3 triggers Ca~(2+) influx and subsequently activates CaMK pathway, however, DAG also activates PKC pathway to increase insulin release.
